The store will be moving to Warwick Road on a ten-year lease.
The news follows the opening of a major outlet in Birmingham city centre at Gateway House on High Street.
A spokesman for Holland & Barrett said: “The Acocks Green premises occupy a busy high street location, which benefits from substantial footfall and a good mix of other quality retailers.
“It represents a significant investment and we are confident it will receive a warm welcome from shoppers.”
The new store is the latest retail addition within the Real Estate Investors’ portfolio.
Holland & Barrett joins existing occupiers including Wilkinson, Boots, Argos, Card Factory and Lloyds Bank.
Ian Clark, senior asset manager at REI, said: “We are delighted at Holland & Barrett’s new venture – this is a further vote of confidence in our portfolio and the strength of the retail marketplace.
“Retail works in the correct environment. There are always challenges to overcome in a highly competitive sector but this deal effectively endorses our on-going asset management strategy.”
